ABSTRACT In order to successfully establish themselves in their natural environment, ferns need habitats with abiotic conditions that are suitable for spore germination and gametophyte development. The objective of this study was to assess the influence of abiotic factors on the initial development of Cyathea phalerata cultivated in vitro. Spore germination and gametophyte development were assessed under varying conditions of surface sterilization, pH, temperature and photoperiod. Exogenous contamination was eliminated by sterilizing spores with 2.5 % NaClO for 15 min and sowing them into a culture medium supplemented with nystatin. Spores germinated at all pHs tested. Background: Genetic and epigenetic changes (DNA methylation) were examined in the tissue-culture propagated interspecific potato somatic hybrids between dihaploid Solanum tuberosum and S. pinnatisectum. Amplified fragment length polymorphism (AFLP) and methylation-sensitive amplified polymorphism (MSAP) were applied to detect the genetic and epigenetic changes, respectively in the somatic hybrids mother plants (1st cycle) and their regenerants (30th cycles sub-cultured). Abstract Cattleya trianae (Linden & Reichb.f., 1860), Colombian national flower, is in danger of extinction due to the destruction of its natural habitats and excessive collection for horticultural purposes. Therefore, in vitro culture is a tool for the conservation of threatened species. Pineapple hybrids selected for their fiber quality are in the phase of agronomic validation in Brazil by the Embrapa Cassava and Fruits research unit. The aim of this study was to evaluate the effect of culture media on in vitro germination and the effect of different substrates on the acclimatization of two Myrciaria tenella types. The study was carried out at the Embrapa Tabuleiros Costeiros Laboratory of Plant Tissue Culture, Aracaju, SE. Cedrela odorata L. is one of the most important timber species currently traded in the Caribbean and Central America; however, it has been intensively exploited. In vitro techniques and clonal propagation can help to develop new plantations and assist in establishing improvement programs for this species. The aim of this study was to develop a protocol to establish in vitro conditions and to micropropagate this species from nodal explants from juvenile cuttings taken from field trees. Micropropagation of Brasilidium forbesii (Orchidaceae) through transverse and longitudinal thin cell layer culture Biological Sciences

An efficient in vitro propagation method was established for Brasilidium forbesii (Hook.) Campacci using transverse and longitudinal thin cell layer (tTCL and lTCL, respectively) culture systems. Six-month-old protocorms from in vitro germinated seeds were used for this study. TCLs (1.0-mm thick) from protocorms were grown on Woody Plant Medium (WPM) supplemented with benzyladenine (BA) (0.5–4.0 µM).The lTCL technique was more efficient for inducing protocorm-like bodies (PLBs) and regenerating shoots than the tTCL technique. Micropropagation of Glandularia perakii Cov. et Schn. (Verbenaceae): a native species with ornamental potential Biocell

Glandularia perakii is a perennial species with beautiful violet flowers that grows in the stony soil of Mendocine pedemont. A plentiful and prolonged flowering confers it an important ornamental potential. In this paper, a method of propagation of G. perakii from nodal segments is reported. Proliferating microshoot cultures were obtained by placing nodal segment on Murashige and Skoog medium (MS) supplemented with 20 g.L-1 of sucrose without growth regulators. In this medium multiplication rate after 20 days was 7.9. Rooted plants were acclimatized successfully.

Microshoots encapsulation and plant conversion of Musa sp. cv. 'Grand Naine' Ciência Rural

Synthetic seed technologies are useful tools for the field delivery of in vitro derived plantlets. In the present study, different encapsulation procedures and their efficacy in the plantlet regeneration using microshoots of banana cv. 'Grand Naine' were evaluated. Two encapsulation systems were evaluated: i) single encapsulation in beads or droplet hardening method; and ii) double layer or hollow beads. The use of different compounds to enhance the capsule conservation and the conversion to plantlets was also evaluated. The conversion capacity was assessed in vitro on water-agar culture medium or in ex vitro conditions with Gerbox® boxes. The single encapsulation system showed 80% conversion. Pigment production and growth of Alternanthera plants cultured in vitro in the presence of tyrosine BABT

The aim of the present study was to investigate the influence of tyrosine on the in vitro growth and the production of the betacyanin pigment in Alternanthera philoxeroides and A. tenella. Nodal segments were inoculated in MS medium containing different concentrations of tyrosine (0, 25, 50 and 75 μM), and the number of sprouts and buds, height, root length, fresh matter of shoots and roots and betacyanin content were evaluated. In A. philoxeroides , the highest production of betacyanin (51.30 mg 100 g-1 FM) was in the stems with the addition of approximately 45 μM tyrosine, while the increase in the leaves was proportional to the tyrosine concentration, and the best average was obtained with a tyrosine concentration of 75 μM (15.32 mg 100 g-1 FM). Somatic embryogenesis and plant regeneration from zygotic embryo explants of onion Horticultura Brasileira

ABSTRACT: The study was undertaken to examine the effect of two synthetic auxins on somatic embryogenesis and plant regeneration from mature embryo explants of 16 onion cultivars. Cotyledons were removed from the embryos and remaining portions were cultured on MS medium supplemented with 1.0 mg/L 2,4-D and 5.0 mg/L picloram alone or in combination (1.0 mg/L 2,4-D + 2.5 mg/L picloram) to produce embryogenic callus. MS medium supplemented with 5.0 mg/L picloram was found to be the best one for both embryogenic callus induction (85%) and callus diameter (3.8 mm).
